It blows the main fuse in the back of the amp.

That looks like it's in series with the output. The 2540 delivers 200W RMS into 2 ohms, so that's 10A RMS. Your replacement would have to pass that JEFF comfortably. You have to ask yourself why it's ...

Given the difference between the two drive channels, it sounds more like something else, if I'm honest.

Can't hurt to borrow a new tube and get a can of cleaner spray to troubleshoot first, anyway. ...
